چکیده

The deoxyribonucleic acid of bacteriophages T2, T4, and T6 contains glucose linked to the hydroxymethyl groups of hydroxymethylcytosine. The work of Sinsheimer, Volkin, Jesaitis, and of Lichtenstein and Cohen (l-4) established the presence of glucosyl-hydroxymethylcytosine residues in the deoxyribonucleic acid and indicated that there is a characteristic glucose to hydroxymethylcytosine ratio for each phage. Lehman and Pratt (5) have now published a detailed analysis showing four types of hydroxymethylcytosine residues which have the following distribution in these phages :

Glucosylation of Deoxyribonucleic Acid II. wGLUCOSYL TRANSFERASES FROM T2- AND TG-INFECTED ESCHERICHIA COLI*t

The deoxyribonucleic acid of the bacterial viruses, T2, T4, and T6, is unique in containing the pyrimidine base, 5-hydroxymethylcytosine, in place of cytosine (2). Attached to a fraction of the 5-hydroxymethylcytosine residues are glucosyl or diglucosyl units in amounts and linkages characteristic for each of the phages. 5-Hydroxymethylcytosine residues with ar-monoglucosyl substituents occur i...

Regulation of the bacterial cell wall: analysis of a mutant of Bacillus subtilis defective in biosynthesis of teichoic acid.

Bacillus subtilis 168ts-200B is a temperature-sensitive mutant of B. subtilis 168 which grows as rods at 30 C but as irregular spheres at 45 C. Growth at the nonpermissive temperature resulted in a deficiency of teichoic acid in the cell wall. A decrease in teichoic acid synthesis coupled with the rapid turnover of this polymer led to a progressive loss until less than 20% of the level found in...

Separation of T-even bacteriophage deoxyribonucleic acid from host deoxyribonucleic acid by hydroxyapatite chromatography.

A simple and rapid method is described for separation of T-even bacteriophage deoxyribonucleic acid (DNA) from host (Escherichia coli) DNA by hydroxyapatite column chromatography with a shallow gradient of phosphate buffer at neutral pH. By this method, bacteriophage T2, T4, and T6 DNA (but not T5, T7, or lambda DNA) could be separated from host E. coli DNA. It was found that glucosylation of t...

The Enzymatic Methylation of Ribonucleic Acid and Deoxyribonucleic Acid V. PURIFICATION AND PROPERTIES OF THE DEOXYRIBONUCLEIC ACID-METHYLATING ACTIVITY OF ESCHERICHIA COLP

The classical investigations of Wyatt (2) and Dunn and Smith (3) established the existence of 5-methylcytosine and 6-methylaminopurine as constituents of the deoxyribonucleic acid of higher plants and animals, and of bacteria and bacterial viruses, respectively. Until recently, the biochemical pathways for the incorporation of these “trace bases” into deoxyribonucleic acid was unknown although ...
